CD10 + Cyclin D1 Double Stain Cocktail Description
Cyclin D1 (also known as bcl-1 or PRAD-1) is a rabbit monoclonal antibody which recognizes a 36kDa protein. Cyclin D1 is a regulatory subunit of certain protein kinases thought to advance the G1 phase of the cell cycle. Due to the rabbit monoclonal technology used in the development of this antibody, its binding capacity is superior to mouse monoclonal antibodies and is virtually background free.
Human CD10, also known as common acute lymphoblastic leukemia antigen (CALLA), has been shown to react with TdT+ lymphoblastic leukemia, follicular germinal cell lymphoma, Burkitts lymphoma, and chronic myelocytic leukemia. It is expressed in some non-lymphoid tissues such as fibroblasts, breast myoepithelium, and brush border of kidney. It does not label mantle cell lymphoma. CD10 is used in a panel for mantle cell lymphoma as a negative marker.
The double stain combination of Cyclin D1 (brown) and CD10 (red) provides both a positive marker (Cyclin D1) and a negative marker (CD10) for mantle cell lymphoma. It is highly suggested that BIOCAREs double stain, CD23 + CD5, be used in tandem. The combination of these four antibodies is the most reliable immunohistochemical panel for mantle cell lymphoma.
